Hibs, who last won the cup in 2016, a side captained by current manager, David Gray (pictured), travel to Dunfermline Athletic managed by their former boss, Neil Lennon.
Spartans will host either Tranent Juniors of Inverness Caley Thistle. Their match was postponed this weekend because of a waterlogged pitch.
Livingston, currently bottom of the Premiership, host St Mirren who are second bottom of the top tier.
Other top ties include last year’s runners-up, Celtic, who travel to Auchinleck Talbot. The East Ayrshire side currently play in the West of Scotland League, Premier Division and have won the Scottish Junior Cup on a record 14 occasions.
Rangers, who have won the trophy on 34 occasions, entertain Annan Athletic who beat Buckie Thistle 4-0 in the third round on Saturday. Annan compete in Scottish League Two.
Aberdeen, the Scottish Cup holders, entertain Raith Rovers.
